Alhydrogel® adjuvant, ultrasonic dispersion and protein binding: A TEM and analytical study

Aluminium-based vaccine adjuvants have been in use since the 1920s. Aluminium hydroxide (alum) that is the chemical basis of Alhydrogel, a widely used adjuvant, is a colloid that binds proteins to the particular surface for efficient presentation to the immune system during the vaccination process. Using conventional TEM and cryo-TEM we have shown that Alhydrogel can be finely dispersed by ultrasonication of the aqueous suspension. Clusters of ultrasonicated aluminium hydroxide micro-fibre crystals have been produced (∼ 10-100 nm), that are significantly smaller than those present the untreated Alhydrogel (∼ 2-12 μm). Introduction The most widely used protocol for the induction of experimental allergic airway inflammation in mice involves sensitization by intraperitoneal (i.p.) injections of the antigen ovalbumin (OVA) used in conjunction with the adjuvant aluminium hydroxide (alum). To evaluate the efficacy of different drug combinations in treating severe gastroesophageal reflux (GER), we studied 80 children with GER. The patients were randomly divided into four groups: group A was treated with domperidone plus magnesium hydroxide and aluminum hydroxide, group B with domperidone plus alginate, group C with domperidone alone, and group D received placebo. At the time of diagnosis and 8 weeks after treatment the patients were clinically evaluated and underwent 24-h continuous esophageal pH monitoring. Experimental study of Si–Al substitution in calcium-silicate-hydrate (C-S-H) prepared under equilibrium conditions.

International audience; C-A-S-H of varying Al/Si and Ca/(Al+Si) ratios have been prepared introducing C-S-H (Ca/Si=0.66 and 0.95) at different weight concentrations in a solution coming from the hydration of tricalcium aluminate (Ca3Al2O6) in water. XRD and EDX (TEM) analyses show that using this typical synthesise procedure, pure C-A-S-H is obtained only for calcium hydroxide concentrations below 4.5 mmol L−1. Otherwise, calcium carboaluminate or strätlingite is also present beside C-A-S-H. The tobermorite-like structure is maintained for C-A-S-H.
